Effects of methamphetamine addiction and withdrawal on serotonin receptor activity

Abstract:

In order to study the reaction of serotonin receptors in depressed humans one must find a suitable model. The use of a psychostimulant withdrawal model in rats is widely recognized as an accurate simulation of the neurochemistry found in depressed humans. This study used such a model in an attempt to determine which serotonin receptor subtypes were affected.

The results of the present study were mixed. Many of the results were similar to those found in previous studies, but the data was unable to further elucidate specific serotonin subtypes that were affected during methamphetamine addiction and withdrawal. The use of fenfluramine (a general serotonin activator) and 8-OH-DPAT (a 5-HT1A-specific agonist) in the present study reaffirmed similar findings using fenfluramine, but failed to identify the 5-HU1A receptor as a useful target of antidepressant drugs. Further study utilizing other challenge drugs may determine which receptors are affected during the fenfluramine treatment.
