Characterization of the AXR4 enhancers in Arabidopsis thaliana

Abstract:

The plant hormone auxin requires import and export proteins for transportation within plants. Mislocalization of the AUX1 auxin import protein is observed in axr4 mutants of Arabidopsis thaliana and results in defects in root growth. The focus of this study is to learn about the functions of the AXR4 gene at the biochemical level by identifying and characterizing axr4 enhancer mutations. In these experiments, 16 potential enhancers were screened on high auxin concentration for root lengths longer than axr4 mutants. Three of these potential enhancers were selected and used in carrying out physiological analysis. Root growth assays revealed increased auxin resistance by WM 20.2.1.1 and WM 2.6.1.2 compared to axr4-3 M30-3. Overall, the enhancers and axr4-3 M30-3 showed delayed gravitropic response, increased root elongation on high auxin concentrations and inhibition of lateral root formation when compared to wild-type columbia.
