Gene expression studies in sour orange and C-22 rootstocks challenged with the nematode, Tylenchulus semipenetrans and the fungus, Phytophthora nicotianae

Abstract:

The cDNA-amplified fragment length polymorphism (cDNA-AFLP) technique was used to identify differentially expressed genes in two citrus rootstocks, sour orange and C-22, when inoculated with the fungus, Phytophthora nicotianae and the nematode, Tylenchulus semipenetrans. Data from cloning and sequencing of eight differential fragments of interest show much homology to the Citrus tristeza virus (CTV) resistance gene locus in Poncirus trifoliata with more than 89% identities and an E value of -75. The profiles generated by AFLP were characterized on two gels, agarose and Poly(NAT)® Elchrom gel and compared for their resolution and clarity. Sharp bands with better resolution were seen on Poly(NAT) when compared to agarose. When the rootstocks were evaluated for the changes in shoot length, shoot weight and root weight when inoculated with Phytophthora and Tylenchulus, sour orange seedlings showed more tolerance to Phytophthora when compared to C-22. These studies will contribute in understanding how plant genes respond to nematode and Phytophthora infections and they may offer a great advantage to citrus rootstock breeding evaluations.
