Catalyst-free-N-ethoxycarbonylation of amino acid esters with diethyl carbonate in aqueous condition

Abstract:

The discovery of safer, more efficient methods for the preparation of chemical products and intermediates is of significant importance to industry, government and academic scientists in the effort to eliminate the economic drain of wasteful practices, as well as the negative impacts to human health and the environment. This study demonstrates a benign and straightforward synthesis of enantio-enriched carbamate derivatives with non-toxic diethyl carbonate and amino acid esters in water. The reactions are carried through at ambient pressure, and can be accelerated by microwave irradiation. The dual nature of diethyl carbonate as reagent and solvent facilitates the separation of the product from the aqueous mixture. The methodology described herein is novel in many regards, namely with respects to the catalyst-free alkoxycarbonylation of amines with diethyl carbonate, the aqueous reaction condition, and the amino acid ester substrate.
