Role of hepatitis C virus F protein in viral protein expression and RNA replication

Abstract:

Programmed ribosomal frameshift is a novel mechanism used by virus to translate protein from 0 reading frame to -2/+1 reading frame. An A-rich slippery sequence and a pseudoknot RNA structure located at 5’end of core coding region constitute a signal for ribosomal frameshift. Hepatitis C virus (HCV) uses such a mechanism to produce F protein from -2/+1 reading frame. However, the function of F is still unclear. In this study, we examined the possible functions of F protein in HCV life cycle. We found that HCV without expressing F protein has poor RNA replication and impaired virus assembly, releasing and infectivity. Our results suggest that F protein has a possible function of regulating RNA replication and maturation process of virus. This observation could give us an idea to develop antiviral therapy.
