Characterization of a box C/D RNA from Haloferax volcanii

Abstract:

In Haloferax volcanii, introns have been identified in pre-tRNATrp and pre-tRNAMet. Both of these tRNAs possess 2'-O-methylation modification at position 34 (Wobble position). The RNA-dependent, 2'-O-methyl modifications are guided by class of snoRNAs (sRNAs in Archaea) called box C/D RNAs. In H. volcanii, pre-tRNATrp intron is known to have box C/D and C'/D' properties, which guide C34 and U39 modification of pre-tRNA Trp. Even though pre-tRNAMet has C34 modification its intron does not exhibit C/D box characteristics. In this study, we report a previously undetected box C/D sRNAs (sR-tMet) from halophilc genomes. We have demonstrated using heterologous reconstitution system that this sRNA can direct 2'-O-methylation of its target residue in the pre-tRNAMet.
